AMPLIFY-201: a lymph node-targeted, mKRAS-specific vaccine in pancreatic & colorectal cancer

In this week's episode of the VJOncoAlert Journal Club, Zev Wainberg, MD, University of California Los Angeles, Los Angeles, CA, discusses results from the Phase I AMPLIFY-201 trial (NCT04853017) of the lymph node–targeted amphiphile vaccine ELI-002 2P in patients with minimal residual pancreatic or colorectal cancer harboring mKRAS mutations. Vaccination induced robust CD4+ and CD8+ T-cell responses, with evidence of antigen spreading to additional tumor-specific targets. Stronger immune responses correlated with improved relapse-free and overall survival. The findings support the potential of mKRAS-targeted vaccines to generate durable immunity and improve outcomes in gastrointestinal cancers.
